ABSTRACT

OBJECTIVE@#To investigate the therapeutic effect of moxibustion on sarcomas from mesenchymal tissues, which have a low response rate to chemotherapy and radiotherapy.@*METHODS@#S180 sarcoma cell line was inoculated in C57BL/6 mice to form transplanted tumor. Moxibustion therapy was directly applied at the transplanted tumor sites, at a distance of 3.0 cm, 10 min per session, till skin temperature reached 45 °C, once a day, for 14 consecutive days of intervention. After the mice were killed, serum was collected and used to detect concentrations of interleukin-10 (IL-10), transforming growth factor-β1 (TGF-β1), IL-4 and interferon-γ (IFN-γ) by Luminex liquid suspension chip. The numbers of Treg@*RESULTS@#Weight of S180 transplanted tumor in the control group was (2.03 ± 0.54) g, and that in the moxibustion group was (1.27 ± 0.29) g, which was statistically different (P = 0.023). The mean value of Foxp3@*CONCLUSION@#Moxibustion may have therapeutic effects on sarcomas by reducing the number of Treg cells in the blood and controlling the infiltration of Treg cells in the TME.

ABSTRACT

Objective In order to meet the national demand for training midwives with higher educational level,our research group aimed to establish cultivation mode of midwifery professional degree for graduate students based on core competence of midwifery.Methods Through literature review and self-designed questionnaire,our research group used the modified Delphi method to conduct two rounds of consultation among 31 experts from six provinces and cities.Results Recovery rates of two rounds of consultation were 88.6% and 100%,authority coefficients were 0.765 and 0.835,Kendall coordination coefficients were 0.296 and 0.303 (P<0.01),and the established cultivation mode of nursing professional degree for graduate students in midwifery contained 2 first-level indicators,9 secondlevel indicators,73 third-level indicators.Conclusion In this study,the experts showed high degree of enthusiasm and authority,reaching a consensus on this issue.The established cultivation system of nursing professional degree for graduate students in midwifery can provide references for training midwifery graduate students in the future.

ABSTRACT

AIM To establish a UPLC method for the simultaneous content determination of glycosides,rutinoside,myricitrin,hyperoside,isoquercitrin,guaijaverin,astragalin,quercitrin and afzelin in the shoots of Toona sinensis Roemer.METHODS The analysis of 60% methanol extract of T.sinensis shoots was performed on a 35 ℃ thermostatic Waters XBridge Shield RP18 column (2.1 mm × 150 mm,1.7 μm),with the mobile phase comprising of acetonitrile-water flowing at 0.35 mL/min in a gradient elution manner,and the detection wavelength was set at 350 nm.RESULTS Eight flavonol glycosides showed good linear relationships within their own ranges (r≥ 0.9992),whose average recoveries were 98.3%-103.5% with the RSDs of 0.46%-3.36%.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of T.sinensis shoots.

ABSTRACT

Objective To develop a magnetic angulometer for the elevated headboard and footboard of manual hospital bed and then evaluate its performances.Methods The angulometer was made of a rectangular PVC,wooden or metal board.The board had a semicircular angle display area at one end,with the straight line at the top while the curve at the bottom of the board.At the other end there was a magnet fixed with a double coated film,which paralleled the top margin of the angulometer.The angle display area was divided into two symmetrical parts,with the angle range from 0 to 90° and the scale being 15°.A string hanging a weight at the center of the angulometer was used to indicate the angle.A control group was formed with the rusults that 14 nurses got with the troditional angulometers in June.2015,and an observation group came into being with the resalts that the 14 nurses obtained with the developed angulometers in July 2015.The two groups with different angulometers were compared on the efficiency for elevating the headboard to some angle appointed.Results The observation group gained significantly higher position compliance with elevation than the control group (x2=75.15,P<0.01),and also statistically higher angulometer efficiency when elevating the headboard to the appointed angle (t=-8.80,-5.68) (P<0.01).Conclusion The angulometer has simple structure and easy operation,and enhances the compliance to body position nursing and the nurse's efficiency.

ABSTRACT

Objective To study the effect of sanguinarine on growth and radiosensitivity of human glioma A172 cells.Methods MTT assay was used to evaluate cell growth.Cell cycle analysis and reactive oxygen species(ROS) burst were performed by flow cytometry assay.Annexin V/PI assay was used to detect cell apoptosis.Colony formation assay was used to detect the influence of sanguinarine on cell radiosensitivity.Results Exposure of A172 cells to sanguinarine led to dose-and time-dependent cytotoxicity with IC50 values of 4.8,3.9 and 3.2 μmol/L for 12,24 and 48 h,respectively.Furthermore,sanguinarine caused an arrest of S phase.After being treated with 5 μmol/L sanguinarine for 24 h,the ratios of early apoptosis,late apoptosis and necrosis were (60.01 ± 3.73)%,(2.70 ± 0.18)% and (3.93 ± 0.76)% respectively in A172 cells compared with the untreated control(t =55.28,8.32,9.51,P <0.05).An increased generation of ROS was found after treatment with sanguinarine,however,NAC inhibited the effect of sanguinarine.As analyzed with multi-target click model fitting curves,the SERD0 of sanguinarine-treated cells was 1.48.Conclusions Sanguinarine inhibits the A172 cells growth via apoptosis induction and ROS burst.Moreover,sanguinarine at a non-cytotoxicity dose increases cell sensitivity to X-rays.

ABSTRACT

Objective To explore the effect of low calorie metabolism on the survival of HeLa cells exposed to X-rays,and the influence of starvation on the antioxidative factors in the blood of rats after irradiation.Methods MTT method was used to evaluate the impact of different concentration glucose on the proliferation of HeLa cells.Colony formation assay was employed to detect the influence of glucose ( 1,5,10 and 25 mmol/L) on radiosensitivity of HeLa cells. Flow cytometry assay was used to analyze distribution of cell cycle and apoptosis.60 male SD rats were randomly divided into 6 groups with 10 rats each.Rats in every two groups were fed ad libitum,fasted for 24 h and fasted for 48 h,respectively.Rats in one group of each approach were respectively exposed to whole-body X-rays at 11 Gy. At 2 h after irradiation,all of rats were sacrificed and their venous blood was collected.Elisa kits were used to detect superoxide dismutase (SOD) and total antioxidant capacity (T-AOC).Results An increased viability was observed in HeLa cells treated with the glucose at low concentration ( <25 mmol/L),while HeLa cell growth was inhibited by glucose at doses of >25 mmol/L. Relevant to cells treated with 1 mmoL/L glucose,SERs (sensitive enhancement ratio) in cells exposed to 5,10 and 25 mmol/L glucose were 1.07,1.10 and 1.23,respectively. A reduction of G2/M and S arrests and apeptosis caused by 6 Gy X-ray irradiation were observed [(49.68 ±1.88)% and (35.54±1.45)% at G2/M phase,(16.88 ±1.22)% and (10.23 ±1.65)% atS phase,t=10.42,5.61,P<0.05]and in the cells treated with 1 mmol/L glucose compared with cells treated with 25 mmol/L glucose [ ( 25.50 ± 0.95 ) % and (7.56 ± 1.07 ) %,t =21.72,P <0.05 ].Without irradiation,calorie restriction exhibited a negligible influence on SOD and T-AOC in rats.However,after 11 Gy irradiation,compared with rats fed ad libitum,the levels of SOD and T-AOC were significantly increased in rats with calorie restriction ( t =40.32,42.78, P < 0.05 ).Conclusions Calorie restriction has a certain radioprotective effect in vivo and in vitro.

ABSTRACT

OBJECTIVES: The present study aimed to investigate the trends in changes in pulmonary function and the risk factors for pulmonary function deterioration in patients with pulmonary tuberculosis after completing treatment. INTRODUCTION: Patients usually have pulmonary function abnormalities after completing treatment for pulmonary tuberculosis. The time course for changes in pulmonary function and the risk factors for deterioration have not been well studied. METHODS: A total of 115 patients with 162 pulmonary function results were analyzed. We retrieved demographic and clinical data, radiographic scores, bacteriological data, and pulmonary function data. A generalized additive model with a locally weighted scatterplot smoothing technique was used to evaluate the trends in changes in pulmonary function. A generalized estimating equation model was used to determine the risk factors associated with deterioration of pulmonary function. RESULTS: The median interval between the end of anti-tuberculosis treatment and the pulmonary function test was 16 months (range: 0 to 112 months). The nadir of pulmonary function occurred approximately 18 months after the completion of the treatment. The risk factors associated with pulmonary function deterioration included smear-positive disease, extensive pulmonary involvement prior to anti-tuberculosis treatment, prolonged anti-tuberculosis treatment, and reduced radiographic improvement after treatment. CONCLUSIONS: After the completion of anti-tuberculosis TB treatment, several risk factors predicted pulmonary function deterioration. For patients with significant respiratory symptoms and multiple risk factors, the pulmonary function test should be followed up to monitor the progression of functional impairment, especially within the first 18 months after the completion of anti-tuberculosis treatment.
